چکیده

In the therapeutic use of penicillin, the question as to whether the minimum effective concentration in vivo is the same, greater, or less than that necessary to kill the same organ'sm in vitro is of obvious importance. Data are here reported with respect to the minimal effective serum concentrations of penicillin in a number of experimental infections in mice and rabbits. As will be shown, these serum levels were usually 2 to 5 times higher than those necessary to kill the same organism in vitro. However, in view of the known concentration differential between the serum and tissue fluids, this suggests that the effective concentration of penicillin at the actual site of infection is in most instances the same as that effective in vitro.

The Effect of the Size of the Inoculum and the Age of the Infection on the Curative Dose of Penicillin in Experimental Infections with Streptococci, Pneumococci, and Treponema Pallidum

The amount of penicillin which suffices to abort pneumococcal and streptococcal infections in white mice and rabbits, as well as syphilitic infection in rabbits, has been shown to increase markedly with the number of organisms inoculated. The curative dose increases also with the age of the infection, presumably owing to the interim increase in the number of organisms in the nfected host. The n...

In vitro culture system to determine MICs and MBCs of antimicrobial agents against Treponema pallidum subsp. pallidum (Nichols strain).

A new procedure for determining the susceptibility of Treponema pallidum subsp. pallidum to antimicrobial agents was developed, utilizing a tissue culture system which promotes the in vitro multiplication of this organism. In the absence of antibiotics, T. pallidum (Nichols virulent strain) multiplied an average of 10-fold when incubated for 7 days in the presence of Sf1Ep cottontail rabbit epi...

Penicillin-binding proteins and peptidoglycan of Treponema pallidum subsp. pallidum.

Penicillin-binding proteins (PBPs) of Treponema pallidum subsp. pallidum (T. pallidum) were characterized by using [3H]penicillin G and a conjugate consisting of ampicillin and 125I-labeled Bolton-Hunter reagent. Both antibiotics specifically radiolabeled proteins with molecular masses of 94, 80, 63, and 58 kilodaltons (kDa); 125I-labeled Bolton-Hunter reagent-ampicillin also radiolabeled sever...

Macrolide Resistance in Treponema pallidum Correlates With 23S rDNA Mutations in Recently Isolated Clinical Strains

BACKGROUND High rates of 23S rDNA mutations implicated in macrolide resistance have been identified in Treponema pallidum samples from syphilis patients in many countries. Nonetheless, some clinicians have been reluctant to abandon azithromycin as a treatment for syphilis, citing the lack of a causal association between these mutations and clinical evidence of drug resistance. Although azithrom...

Mitogenic responses of hamsters infected with Treponema pertenue Lack of correlation with passive transfer of resistance.

Infection of the CB/Ss Lak hamster with Treponema pertenue is characterised by chronic cutaneous lesions and lymph nodes teeming with treponemes. Throughout the course of infection lymph node and spleen cells responded poorly to the mitogens concanavalin A, phytohaemagglutinin, and lipopolysaccharide. This impairment preceded clinical signs of infection and correlated well with the chronicity o...
